This was listed in the job posting:  You will need to successfully complete a background security investigation before you can be appointed into this position. 

What will they be checking and what is the process for this kind of investigation?

They will be checking your credit, past employers, neighbors, family, friends, and review of finances.

What kind of things can cause you to "fail"?    Do they do drug testing, polygraphs, medical checks?

The extent of the background investigation depends on the type of position.  The most basic background investigation will include an FBI fingerprint check for criminal records and a local law enforcement.  If you are male, selective service registration will be verified.  And you will have to provide acceptable proof of US citizenship.  More sensitive positions can require reference checks, previous employers, education institutes, your neighbors, drug tests, polygraphs, credit reports, personal interviews, follow-up on developed leads, etc.  The most important thing to remember is to complete the forms for position with total honesty because omission of even a minor item can be grounds for disqualification.  Conversly, and individual with a less than ideal record can still be hired if they disclosed thier complete record.
 
The employing agency can tell you exactly what type of investigation is required and you can then Google it for details
